Abstract:

The knowledge related to the methodology of the Field trial study as a type of intervention studies, yet for many of our researchers is not fully understood. The aim of the current study was a better understanding of conducting this type of research. Field trial studies are done on healthy individuals and aim to prevent. These types of studies such as clinical trials are performed on both individual and collective levels. One type of these studies is Community Intervention trial which is usually done on a large scale population. Field trial study should be carried out in stages, such as the formulation of hypotheses, selection of the population (reference population, study population, and sampling), measuring the baseline variables (before conducting preventive intervention), random allocation of subjects to intervention and control groups, doing interventions and measuring outcome. The methodology of Field trial studies is very similar to clinical trials. The difference is that Field trials are conducted on healthy individuals and aim to prevent and also the sample size required to this type of study is relatively more, and these studies are usually time consuming and costly.

Abstract:

Abstract:

Background & Aims: Urinary tract infection (UTI) is one of the most frequently acquired bacterial infections caused by a large genetically heterogeneous group of Escherichia coli, which are called uropathogenic E. coli (UPEC). Cystitis and pyelonephritis are two most common symptoms seen in patients with UTI. The genetic diversity of this organism has hampered the identification of UTI strains and it is unclear whether all UPEC isolates are capable of causing both cystitis and pyelonephritis. Therefore, Careful selection of appropriate genotyping methods is mandatory. The most popular method is Pulsed Field Gel Electrophoresis (PFGE) that is used in the present study to evaluate the genetic patterns of UPEC. Methods: In this cross-sectional study a total of 90 E. coli strains consisting of 48 isolates causing pyelonephritis and 42 isolates causing cystitis in children were analyzed by PFGE and their corresponding patterns were compared. Results: Sixty six PFGE profiles were obtained from the genome of E. coli strains by this genotyping method. Most strains exhibited twelve and thirteen bands and the patterns with eight or nineteen bands had the lowest rate. Genome size of strains was between 1610-4170 kbp. Conclusion: According to these results, it can be suggested that in some cases the strains causing pyelonephritis or cystitis have common patterns and different clinical symptoms could be attributed to different gene factors.

Abstract:

THE MOST ECONOMIC AND ENVIRONMENTALLY SAFEST STRATEGY FOR CONTROLLING POTATO LEAFROLL VIRUS (PLRV) IS THE USE OF RESISTANT POTATO GENOTYPES. IN THIS STUDY, USING A RANDOMIZED COMPLETE BLOCK DESIGN IN A Field trial, THE INFECTION RATES OF 12 POTATO GENOTYPES TO PLRV WERE EVALUATED. EACH PLOT OF THE DESIGN CONSISTED OF FIVE PLANTS OF EACH POTATO GENOTYPE OR CULTIVAR. INOCULATION OF TEST PLANTS WITH THE VIRUS WAS DONE BY PUTTING 10 PLRV-CARRYING APHIDS ON EACH TEST PLANT. THE APHIDS WERE THEN KILLED BY INSECTICIDE APPLICATION. ONE MONTH LATER USING BOTH SYMPTOM DEVELOPMENT AND THE ENZYME-LINKED IMMUNOSORBENT ASSAY (ELISA) TEST THE STATUS OF TEST PLANTS REGARDING TO INFECTION WITH PLRV WERE EVALUATED. SIGNIFICANT DIFFERENCES WERE FOUND BETWEEN THE PLRV-INFECTION RATES OF THE POTATO GENOTYPES TESTED. THE GENOTYPE 803970/13 WAS EVALUATED AS HIGHLY RESISTANT TO PLRV. THE CULTIVAR SANTE WAS RESISTANT AND THE CULTIVAR LADY ROSETTA AND THE GENOTYPE 397015/31 WERE MODERATELY RESISTANT. THE REST OF GENOTYPES OR CULTIVARS WERE MODERATELY SUSCEPTIBLE, SUSCEPTIBLE OR HIGHLY SUSCEPTIBLE. MOREOVER, A SIGNIFICANT CORRELATION (79%) BETWEEN THE INFECTION RATES OF THE TEST PLANTS BASED ON SYMPTOM DEVELOPMENT AND THOSE OF THE ELISA TESTS WAS FOUND.

Abstract:

Potato leaf roll disease is one of the most important and widely distributed viral diseases of potato. Like other plant viruses, the use of resistant cultivars is the most effective control measure of this disease. In this study, the reactions of some potato cultivars and genotypes to Potato leafroll virus (PLRV) were evaluated in a Field trial experiment. The experiment conducted as a randomized complete block design with 12 treatments and three replications. Each plot of the experimental design included a planting row of five potato plants of each cultivar/genotype. The experimental plants were then inoculated with the virus by putting 10 PLRV-carrying green peach aphids on each plant. One month after inoculation, the plants were examined for PLRV infection by observing symptoms development and using enzymelinked immunosorbent assay (ELISA) test. The results revealed significant differences between the PLRV-infection rates of the potato cultivars/genotypes tested. The potato genotype 803970/13 with having no infected plant was evaluated as highly resistant to PLRV. The cultivar “ Sante” was resistant, the cultivar “ Lady Rosetta” and the genotype “ 397015/31” were moderately resistant, cultivar “ Diamant” was moderately susceptible and the rest of genotypes or cultivars were found susceptible or highly susceptible. The results also showed a significant correlation (79%) between the infection rates of the test plants based on symptom development and those of the ELISA tests.

Abstract:

Background and Aim: Sexuality education is one of the most important educational needs of adolescents, and mothers can have a key role in sex education of their daughters. This study aimed at developing, implementing and evaluating a skill-based training program for mothers for sexuality education of adolescent girls in Mahshahr, Iran.Materials and Methods: This was a Field trial conducted on 140 mothers having 12- to18-yearold girls between September and March 2012. Data were collected using two reliable questionnaires developed by the investigators ─ a "Parent-Child Relationship: Mother" questionnaire for assessing the mothers’ communication skills completed by the adolescent girls, and a "Knowledge and attitude about sexuality education of adolescents" questionnaire for assessing the mothers’ knowledge and attitude about sex matters and communication, skills completed by the mothers themselves. Based on the results, an educational intervention for mothers was designed consisting of three training sessions and implemented. The mothers’knowledge, attitude and communication scores after a period of three months were compared with the respective baseline scores. Results: Three months after the intervention, the mean scores of mothers’ knowledge and attitude about sexuality matters increased significantly (in both cases p<0.01). However, there was no statistically significant difference between the initial and final scores of mothers’ communication skills (p=0.37).Conclusion: Based on the findings it can be concluded that the designed educational program was effective in improving the knowledge and attitudes of the mother about sexuality matters although it did not affect their communication skills. Promoting communication skills of mothers as an important factor for sexuality education of their daughters requires regular, continuous educational sessions, as well as practice and experiences.
